Men genetically predisposed to male-pattern baldness face a 7% increased risk of hair loss when using GLP-1 drugs like Ozempic, Wegovy, and Zepbound, according to a study published in the Journal of Investigative Dermatology. The research, led by NYU Langone Health, identifies a genetic link between GLP-1 receptor activity and androgenetic alopecia, suggesting potential for pre-prescription screening and combination treatments.

A new study has identified a genetic connection between GLP-1 receptor activity and male-pattern hair loss, revealing that men already at risk for androgenetic alopecia face a 7% increased chance of hair loss when using medications like Ozempic, Wegovy, and Zepbound. The research, published in the Journal of Investigative Dermatology, analyzed genetic data from over 200,000 individuals and found that variants linked to higher GLP-1 receptor protein levels were more common in men with inherited hair loss.

Genetic Link Confirmed

The study, led by Lynn Petukhova, PhD, an assistant professor in the Ronald O. Perelman Department of Dermatology and the Department of Population Health at NYU Grossman School of Medicine at NYU Langone Health, focused on the GLP1R gene, which regulates GLP-1 receptor proteins. Researchers compared genetic patterns associated with GLP1R activity to those linked to androgenetic alopecia, finding that men with higher GLP-1 receptor activity had a 7% greater risk of hair loss. This association persisted even after accounting for hypertension, a known contributor to hair thinning, according to the study.

Our study provides the first genetic link between GLP-1 use and an increased risk of male- pattern hair loss from androgenetic alopecia, an association long suspected but until now not shown scientifically, Petukhova said in a statement reported by ScienceDaily.

Methodology and Key Findings

Researchers used large genetic databases, including the eQTLGen database (31,684 participants) and the Complex Traits Genetics group (205,327 participants), to examine the relationship between GLP-1 receptor activity and hair loss. The study employed Mendelian randomization, a technique that uses genetic variants as natural experiments to infer causality. This approach helped isolate the effect of GLP-1 receptor activity from other factors like rapid weight loss, which is often cited as a cause of hair shedding in GLP-1 users.

While the study confirmed a 7% increased risk, it did not establish a direct causal relationship between GLP-1 drugs and hair loss. Instead, the researchers suggest that the genetic overlap between GLP-1 receptor activity and androgenetic alopecia indicates a shared biological pathway.

Potential Clinical Implications

The study’s authors propose that future research could lead to genetic screening for patients prescribed GLP-1 medications, allowing doctors to identify individuals at higher risk of hair loss. Our findings suggest that, if future experiments prove successful, some men, and possibly women too, could be screened and benchmarked for their risk of hair loss before being prescribed GLP-1 medications, Petukhova said according to Knowridge.

However, the study’s findings are limited to men, as male-pattern hair loss has been more extensively studied than female-pattern baldness. We also plan to investigate whether the same genetic connection found in men is present in women, Petukhova noted in an interview with Earth.com. Additionally, the genetic databases primarily included individuals of European ancestry, raising questions about the results’ applicability to more diverse populations.

Hair Loss and Weight Loss: Understanding the Connection with Ozempic and Wegovy

The research also underscores the importance of distinguishing between hair loss caused by rapid weight loss and that potentially linked to GLP-1 receptor activity. Hair thinning has been reported by both men and women taking GLP-1 drugs. Until now, researchers have generally attributed much of that shedding to rapid weight loss, since these medications lower blood sugar and reduce appetite, reported IndicaneWS. The study’s genetic approach provides a clearer picture of the biological factors at play.

While the 7% risk is modest, the findings add to the growing body of evidence linking GLP-1 drugs to unintended side effects.
