The authors suggest that these findings could represent a step toward the clinical translation of klotho as a rejuvenating treatment for brain function.

Klotho is a longevity protein that declines with age and has been shown to extend the lifespan of mice; it has also recently been found to improve synaptic function and enhance cognitive function in mice.

Cognition is a key brain function that deteriorates with aging and age-related diseases, such as Alzheimer’s disease. With the aging of the world population, cognitive defects have become a biomedical challenge that requires effective pharmacological interventions.

To explore whether klotho has similar effects on cognition in non-human primates, Dena Dubal’s team from the University of California-San Francisco (USA) administered a single low dose of klotho (10 micrograms per kilogram of body weight) to 18 elderly macaques (average age approximately 22 years).

The results show that a single dose of klotho leads to an improvement in cognitive function in older primates, as measured by tests assessing working and spatial memory, and that these improvements persisted for at least two weeks. Higher doses did not show any cognitive improvement.

The authors conclude that the beneficial effects of klotho on cognition extend to non-human primates. These findings could help in the future development of an intervention to counteract cognitive decline in older people.
