Susumu Tonegawa, the Nobel Prize-winning molecular biologist who transformed our understanding of the immune system and memory storage, died on July 11 at age 86. His death was announced this week by the Massachusetts Institute of Technology, where he served as a professor and belonged to the Picower Institute for Learning and Memory.
Solving the Puzzle of Antibody Diversity
Tonegawa’s scientific career was defined by two distinct phases. In the 1970s and early 1980s, he tackled one of biology’s most persistent questions: how the human body generates a vast, specialized repertoire of antibodies to combat an infinite variety of pathogens.
Through his research, Tonegawa demonstrated that B cells do not carry a separate gene for every possible antibody. Instead, he discovered that these cells assemble antibody genes by recombining specific segments—a process now known as V(D)J recombination. This breakthrough, which explained the mechanism behind antibody diversity,
earned him the 1987 Nobel Prize in Physiology or Medicine. He was the first Japanese recipient of the honor.
A Second Act in Neuroscience
After cementing his legacy in immunology, Tonegawa transitioned to neuroscience, applying the same genetic rigor to the study of the brain. At MIT, he focused on the physical basis of memory. His laboratory identified memory engrams,
which are ensembles of cells that encode episodic memories. His team demonstrated that these memory traces could be activated or altered in mice, providing a new framework for understanding how the brain stores and retrieves information.
Leadership and Legacy at the Picower Institute
Beyond his individual discoveries, Tonegawa was instrumental in building the research communities that would carry his work forward. He built the laboratory community that became MIT’s Picower Institute for Learning and Memory and also led the RIKEN Brain Science Institute in Japan. Colleagues noted that his influence extended far beyond his published papers, as he mentored generations of scientists who continue to study engram research today.
