A research team at the University of Arkansas for Medical Sciences has discovered a drug to prevent Alzheimer’s disease in people with the APOEε4 gene associated with the disease.

Between 50 and 65% of people with Alzheimer’s have inherited the APOEε4 gene, significantly increasing their risk of developing the disease, those with one copy of APOEε4 have a three-fold increased risk, while those with two copies have a risk between 12 and 15 times higher. This drug candidate, CBA2, targets the harmful effects of the inherited APOEε4 gene.

Research by the team has revealed new insights into APOEε4-related drug targets and damaging mechanisms that contribute to Alzheimer’s disease. This series of discoveries is considered groundbreaking in the field of neuroinflammation and Alzheimer’s research.

Balasubramaniam’s unique computational modeling and analysis enabled the rapid detection of around 800,000 compounds in just two days. The results of computer-simulated pharmacological actions targeting ApoE4 were validated in various in vitro and in vivo model systems.

The team aims to conduct further preclinical investigations of the drug candidate CBA2 and explore other potential candidates for Alzheimer’s prevention. They hope that people who carry APOEε4 will one day be able to reduce their risk of Alzheimer’s by taking this medication regularly.

The research findings were published in Communications Biology, with additional information available in Alzheimer’s and Dementia and the Journal of Neuroinflammation.
