Researchers at Emory University have discovered that daily intake of 5,000 IU or more of vitamin D is linked to a 13% higher score on cognitive tests among older adults experiencing sleep disturbances and mild cognitive impairment, according to a study published in Sleep Medicine.
With more than 7 million Americans currently living with Alzheimer’s dementia—a figure projected to nearly double to nearly 13 million by 2050—scientists are racing to identify accessible, modifiable risk factors according to research from Emory University.
Targeting Early Cognitive Decline and Sleep Disturbances
The study evaluated 54 adults aged 50 or older who either had diagnosed or suspected mild cognitive impairment (MCI) alongside documented sleep disturbances. Investigators used the Montreal Cognitive Assessment (MoCA) to screen memory and thinking skills, while sleep quality was measured using the Pittsburgh Sleep Quality Index (PSQI) and the Epworth Sleepiness Scale (ESS).
When researchers analyzed self-reported vitamin D intake against these cognitive and sleep metrics, a striking pattern emerged. Participants taking a daily dosage of 5,000 IU or more scored roughly 13 percent higher—an average of 3.78 points higher—on the MoCA cognitive screening tool compared to participants who reported taking no vitamin D supplements. This statistical association held firm even after adjusting for potential confounding variables like age, sex, body mass index, and education level.
Lower daily doses did not yield statistically significant score improvements. Furthermore, the form of the supplement appeared to make no difference: participants taking vitamin D2, typically sourced from plants or fungi, performed similarly to those taking animal-derived or sun-synthesized vitamin D3 forms.
A Critical Window for Brain Health Intervention
Mild cognitive impairment acts as a clinical bridge between normal cognitive aging and dementia. Researchers emphasize that catching cognitive shifts during this transitional phase may offer the best chance to protect neurological function before damage compounds.

Sleep disruption is a well-documented complication in neurodegenerative conditions. Approximately 50 percent of individuals with moderate to severe Alzheimer’s disease experience sleep disturbances, pointing to a complex two-way relationship where poor sleep cycles and cognitive decline feed into one another as detailed by Emory researchers. Because vitamin D receptors influence muscle function, nerve signaling, and sleep-wake regulation, deficiencies have frequently been tied to insomnia and nighttime awakenings.
Cautious Interpretation and Next Steps for Vitamin D Research
Despite the encouraging correlation, study authors urge significant caution. Because this investigation was strictly observational, it establishes an association rather than direct causation, meaning researchers cannot yet claim that vitamin D directly enhances cognitive performance or prevents disease progression.

Additionally, a daily intake of 5,000 IU sits slightly above the maximum recommended daily upper limit of 4,000 IU set to avoid hypercalcemia according to safety guidelines. The research team emphasizes that larger, randomized clinical trials are required to confirm these preliminary findings, evaluate potential toxicity risks, and determine whether targeted supplementation can safely alter the trajectory of cognitive decline.
