Washington University Researchers Reveal Angelman Syndrome Stress Cause

Researchers at Washington University School of Medicine revealed that even a moderate 25% reduction in UBE3A triggers a false cellular stress response by causing asparagine synthetase to accumulate, stunting brain growth and impairing protein synthesis in developing neurons.

Enzyme Accumulation Drives Angelman Syndrome

Yet the precise molecular chain reaction that stalls brain development remained opaque. Investigators at Washington University School of Medicine in St. Louis set out to map that trajectory by generating cell models lacking a functional UBE3A gene.

Among those candidates, a single metabolic enzyme stood out: asparagine synthetase, known as ASNS. Responsible for constructing the amino acid asparagine for general cellular protein assembly, ASNS is normally benign under standard physiological conditions. But in this developmental disorder, its uninhibited accumulation alters normal cellular chemistry without causing direct physical damage to membranes or organelles.

The study, published in Nature, is titled Persistent neuronal stress signaling underlies Angelman syndrome phenotypes. Its authors include Kwang-Soo Kim, Cory Gardner, Alissa Cullen, Jalin A. Stelzer, Kevin Cho, Ashish Sharma, Mati T. Nemera, Travis Law, Huma Naz, Guoyan Zhao, Harrison W. Gabel, Gary J. Patti, Erik S. Musiek, Tae-Hyuk Ahn, and Jason J. Yi.

Excess Asparagine Synthetase Triggers False Neuronal Stress

The discovery that ASNS overabundance drives pathology rather than merely marking it offers a fresh paradigm for neurodevelopmental arrest. Instead of acting through direct cellular toxicity, the excess enzyme serves as an aberrant biochemical trigger that signals depletion.

The neurons are tricked into thinking that they’re in stress when they’re not — that’s bad because they will then not grow normally.

This false signal activates an integrated stress response within the brain, prompting developing neurons to conserve cellular resources by curtailing protein synthesis. Proteomic evidence and substrate specificity confirm that this 25% reduction in UBE3A is enough to alter ASNS abundance, suggesting that neuronal proteostasis is finely tuned to dosage changes. Detailed molecular analyses show that ASNS overabundance activates stress signaling and causes a global reduction in neuronal protein synthesis, giving the work more depth than a simple stress is up, translation is down narrative.

For proteasome inhibition assays, cells were treated with 15 micromolar MG132 for 12 hours before collection, while control cells were treated with an equal volume of dimethyl sulfoxide as a vehicle control. Primary cortical neurons were cultured and maintained using the B-27 Plus neuronal culture system supplemented with N2 and penicillin-streptomycin, with half of the neuronal culture medium replaced every 2 days until the desired day of the experiment.

Genetic Correction and Behavioral Recovery in Mouse Models

To test causality rather than mere correlation, researchers utilized mouse models maintained on a C57BL6/J background. Protocols for animal use were reviewed and sanctioned by the Institutional Animal Care and Use Committee of Washington University in St. Louis, with subjects housed in a pathogen-free vivarium overseen by the Division of Comparative Medicine at the Washington University School of Medicine, holding full AAALAC International accreditation. Angelman syndrome models were produced by breeding heterozygous females with wild-type males, whereas an Asns-heterozygous subject was created at the Washington University Mouse Genetics Core by applying a guide RNA to delete a single nucleotide at the Q17 codon within exon 2 of Asns. Mouse holding rooms were maintained in a controlled environment with an ambient temperature of 20 to 24 degrees Celsius and a relative humidity of 40 to 60%, and cages were furnished with 160 grams of corn-cob bedding, 220 grams of food pellets, and 380 milliliters of water. After 5 days of acclimation, sleep-wake states were recorded without disturbance over 7 days using PiezoSleep.

The genetic rescue experiment tested causality by reducing ASNS overabundance toward normal levels. Blocking this downstream enzyme restored normal neural protein synthesis, normalized brain growth, and reversed behavioral deficits in mouse models.

Visible enhancements in brain weight, microcephaly, and multiple behavioral metrics—such as marble burying, hindlimb clasping, and time spent in the center of an open field—were documented in the resulting rescue mice.

We still don’t know a whole lot about the disease. We haven’t been able to identify all of the things that the missing gene is doing and rescue the things that it is not doing in Angelman’s syndrome. But I think our data are the most complete so far to begin answering those questions.

The findings indicate that targeting ASNS and the stress response could hold therapeutic potential in Angelman syndrome, reframing the condition as a disorder of persistent neuronal stress and impaired translational homeostasis.

Which missing gene functions have yet to be identified, and what prevents researchers from rescuing all of them in Angelman syndrome?
