Boulder biotech Enveda announced in August 2026 that its experimental oral drug ENV-308, designed to mimic the exercise-induced hormone Lac-Phe, successfully cleared a Phase 1 clinical trial in 88 healthy adults. Researchers reported an exceptional gastrointestinal safety profile, setting the stage for Phase 2 trials testing weight maintenance after GLP-1 therapy.

Enveda revealed that its experimental compound, ENV-308, has cleared its initial safety evaluation in humans. How ENV-308 Mimics the Chemistry of Exercise

The drug targets N-lactoyl-phenylalanine, commonly known as Lac-Phe. Stanford University researchers first discovered the molecule in 2022, finding that the body produces it naturally when lactate—released by active muscles during vigorous exercise—combines with the amino acid phenylalanine. Once in the bloodstream, Lac-Phe signals the brain to dial down appetite. However, naturally occurring Lac-Phe clears the body far too quickly to serve as an effective standalone treatment for weight control.

Enveda used its proprietary artificial intelligence platform to engineer ENV-308 as an orally bioavailable analog intended for daily dosing designed to reproduce the effects of Lac-Phe. Phase 1 Safety Data and the Search for a GLP-1 Alternative

During the Phase 1 trial, Enveda administered ENV-308 at varying strengths to 88 healthy adult volunteers. Because early-stage trials focus primarily on safety and tolerability rather than efficacy, the experiment was not designed to measure weight loss. Instead, researchers tracked adverse events and pharmacokinetic markers across the full dose range.

Participants tolerated the treatment well, with no serious adverse events, discontinuations, or dose interruptions reported. Furthermore, the company highlighted an “exceptional” gastrointestinal safety profile. That distinction matters significantly in the obesity treatment landscape, where popular GLP-1 receptor agonists like Wegovy and Zepbound frequently trigger nausea, vomiting, and other gut-based side effects that prompt some patients to discontinue use.

Biomarker Findings and the Road to Phase 2 Trials

Although the initial trial did not evaluate weight outcomes, researchers tracked circulating leptin as an exploratory pharmacodynamic marker. Buoyed by these preliminary findings, Enveda plans to launch a Phase 2 trial. This next phase will directly investigate whether an oral Lac-Phe mimetic can maintain weight loss after patients withdraw from GLP-1 therapy. Preclinical data also suggest the compound could potentially be used alongside existing therapies, possibly allowing patients to reduce dosing frequency while maintaining tolerability according to company disclosures.

Clinical investigators emphasize that while the early safety data provide an encouraging foundation, comprehensive human trials are still required to determine whether ENV-308 can effectively influence body composition and weight in everyday patients.
