Innovative Cancer Treatment Shows Promise for Head and Neck Squamous Cell Carcinoma Patients

A groundbreaking new approach combining pulsed electrical field ablation with intratumoral immunotherapy is demonstrating significant efficacy and safety in treating advanced head and neck squamous cell carcinoma (HNSCC), according to data presented at the 2025 Society for Immunotherapy of Cancer (SITC) conference.The Williams Cancer Institute (WCI) announced the positive findings from its study, titled “Pulsed electrical field ablation plus intratumoral immunotherapy for head and neck squamous cell carcinoma.” The research offers a potential lifeline for patients with arduous-to-treat tumors who may not be eligible for customary therapies.

“Our findings reinforce the potential of combining precision-based local ablation with intratumoral immunotherapy to activate an immune response against cancer,” stated Dr. Jason Williams, Founder and Director of Interventional Oncology and Immunotherapy at WCI. “The data show that this approach is safe and effective for treating head and neck squamous cell carcinoma and may offer new hope for patients with difficult-to-treat tumors who are not candidates for conventional therapies. Addressing the Challenges of HNSCC

Head and neck squamous cell carcinoma is a common and often aggressive cancer frequently linked to tobacco use or human papillomavirus (HPV) infection. These tumors are often diagnosed at advanced stages, making treatment complex. Traditional methods like surgery, chemotherapy, and radiation can be challenging due to the tumor’s location and complexity.

The WCI study focused on a single patient with advanced HNSCC who underwent a novel treatment regimen. This involved combining pulsed electrical field ablation – a minimally invasive procedure using electrical pulses to destroy tumor cells – with a series of intratumoral immunotherapy injections directly into the tumor.

The Treatment Protocol

The patient’s treatment plan consisted of the following key components:

  • Four cycles of pulsed electrical field ablation were administered during the initial session to disrupt the tumor cells.
  • Intratumoral injections containing an immunotherapy mixture, iodinated contrast for enhanced imaging, and gelfoam slurry for localized retention were delivered.
  • Follow-up immunotherapy injections were administered at weeks three and four, with additional injections given again at the eight-month mark post-treatment.

Remarkable Results and Durable Response

The innovative approach yielded substantial tumor regression and a sustained clinical response.Importantly, no treatment-related toxicity or procedural complications were observed. Imaging at 14 months revealed significant radiologic and immunologic elimination of the tumor. Remarkably, by 22 months, the patient’s previously disfiguring mass had completely resolved.

The study, authored by Erik Soule, Carlos A vargas, Eduardo Cortes, Nathan Goodyear, and Jason R Williams, has been published in the Journal for ImmunoTherapy of Cancer and is available for further review.
