San Antonio, TX, December 12, 2025 – A new treatment approach for a common form of breast cancer is showing promising early signs of extending life, even if it doesn’t promptly slow tumor growth. Sacituzumab govitecan (SG) demonstrated a trend toward improved overall survival in patients with hormone receptor-positive, HER2-negative metastatic breast cancer, according to results unveiled at the 2025 San Antonio Breast Cancer Symposium.
ASCENT-07: A Closer Look at the Trial Design

The phase 3 ASCENT-07 trial (NCT05840211) enrolled 690 adults diagnosed with hormone receptor-positive, HER2-negative, locally advanced or metastatic breast cancer. Participants where randomly assigned to receive either SG at a dose of 10 mg/kg intravenously, or chemotherapy chosen by their physician – specifically, capecitabine, nab-paclitaxel, or paclitaxel. The study accounted for prior treatment with CDK4/6 inhibitors, HER2 status, and geographic region during randomization.2
The primary goal of ASCENT-07 was to measure progression-free survival (PFS), meaning the time until the cancer started to grow again, as assessed by autonomous experts using standardized criteria (RECIST v1.1). Researchers also tracked overall survival (OS),objective response rate (ORR),and quality of life,along with duration of response and potential side effects.2
Initial Findings: A Mixed Picture
While SG did not considerably improve progression-free survival compared to chemotherapy, the study revealed some encouraging results. The objective response rate was 32.4% with SG versus 21% receiving chemotherapy. Despite the higher rate of severe side effects with SG, dose reductions were comparable between groups, and fewer participants discontinued treatment with SG.2
What Does This Mean for Patients?
ASCENT-07 did not achieve its primary goal of demonstrating a statistically significant betterment in progression-free survival when assessed by independent review. though, the study revealed no new safety concerns. The early indication of a potential overall survival benefit with SG,coupled with its manageable tolerability,suggests further examination is warranted.
These findings indicate that while SG may not dramatically extend the time before a cancer progresses in this setting, its potential to improve survival and its acceptable side effect profile make it a promising avenue for continued research.
