The ongoing impact of COVID-19 extends far beyond the initial acute illness, with a significant number of individuals experiencing persistent symptoms—a condition commonly known as long COVID. While researchers continue to unravel the complexities of this condition, a new study suggests that vitamin D3 supplementation may offer a potential avenue for reducing the risk of developing long COVID, though it doesn’t appear to impact the initial stages of the infection. This finding adds to the growing body of research exploring accessible interventions to mitigate the long-term effects of the virus.
Published online March 12 in the Journal of Nutrition, the research, led by Davaasambuu Ganmaa, M.D., Ph.D., of Brigham and Women’s Hospital and Harvard Medical School in Boston, investigated whether vitamin D3 could influence the course of COVID-19 and the likelihood of developing lingering symptoms. The study focused on adults newly diagnosed with COVID-19 during a period when vaccination rates were still evolving and treatments were limited. The question of long COVID and potential preventative measures remains a critical area of investigation.
Study Design and Key Findings
The study employed a cluster-randomized, controlled trial design. This means that participants, along with one cohabiting contact, were randomly assigned to receive either a high dose of vitamin D3 or a placebo. A total of 863 participants received vitamin D3 (a loading dose of 9,600 IU/day for two days, followed by 3,200 IU/day for four weeks), while 884 received a placebo. Researchers then tracked health care utilization and other clinical outcomes over a four-week period.
The primary outcome—the four-week incidence of health care utilization—showed no significant difference between the two groups. Specifically, 28% of the vitamin D3 group and 29% of the placebo group required health care intervention (odds ratio, 0.97; 95% confidence interval, 0.75 to 1.24). Other pre-specified outcomes related to the acute phase of the illness also did not differ significantly. However, a closer look at the data revealed a potentially important trend.
In a per-protocol analysis—meaning the analysis focused on participants who adhered to the study protocol—researchers observed a non-significant trend toward a reduction in the prevalence of long COVID at eight weeks among those who received vitamin D3 (odds ratio, 0.78; 95% confidence interval, 0.59 to 1.03). This suggests that vitamin D3 supplementation *may* reduce the risk of developing long COVID, but the evidence isn’t yet conclusive.
The Vitamin D and Immune Function Connection
